Day Zero Diagnostics (DZD) is an exciting startup revolutionizing infectious disease diagnostics, by leveraging cutting-edge sample prep technologies, whole genome sequencing and machine learning. We are building the next generation of IVDs, able to perform comprehensive bacterial species ID and antimicrobial resistance and susceptibility (AMR/S) profiling in less than 8 hours of sample receipt, without the need for culture.
Our first application is for Sepsis. Sepsis is responsible for about a third of hospital deaths and costs hospitals in the US about $24B annually. Using the current culture-based approach for pathogen ID and AST that takes 2-5 days and has a 40% failure rate, patients with Sepsis are treated with broad-spectrum antibiotics, leading to significant toxicity, higher rates of organ injury, increased risk of c. difficile infection, and contributing to the growth of the antibiotic resistance problem, globally.
By providing an accurate and comprehensive diagnosis within the first cycle of treatment, patients can get appropriate antibiotic therapy for systemic infections such as sepsis, reducing hospital treatment durations and costs.

SUMMARY
Day Zero Diagnostics is seeking a Laboratory Technician II to support the Sequencing Technologies team. This candidate is responsible for daily high-throughput sample processing, conducting DNA library preparation, quality control (QC) assessments, and whole-genome sequencing (WGS) on both short- and long-read platforms with minimal guidance. They will handle reagent preparation and project-specific inventory while strictly following SOPs, meticulously documenting processes and delivering high-quality, timely results.
Located in DZD’s new, state-of-the-art facility in Watertown, MA, this role provides opportunities for developing skills surrounding training, logistics, and data analysis in addition to wet lab development.
RESPONSIBILITIES
- Perform DNA library preparation, QC assessments including TapeStation and fluorescent quantitation assays, and WGS for diagnostic workflows on short-read and long-read sequencing platforms with minimal guidance
- Execute repetitive NGS workflows and strictly follow SOPs for specimen handling, processing, and reporting
- Collaborate with interdisciplinary teams and integrate NGS workflows into broader research initiatives
- Identify risks/issues that may adversely affect test performance and notify the appropriate supervisor
- Maintain meticulous documentation on sample processing and tracking, QC checkpoints, and any corrective actions taken when tests deviate from the laboratory’s established SOPs
- Train/mentor wet-lab team members, provide guidance on SOPs, techniques and workflow improvements
- Conduct routine maintenance of instruments, manage inventory, and order essential reagents and consumables
- Prepare and communicate SOPs, data summaries, technical reports, and presentations for internal use
QUALIFICATIONS
- A bachelor's degree in a chemical, physical, biological, or clinical laboratory science from an accredited institution, or a 4 year medical or biotechnology degree, is required to fulfill CLIA requirements.
- 1-2 years of wet lab experience in academic and/or industry setting
- Experience with laboratory methods/techniques associated with NGS pipelines, including Illumina-based DNA library construction, QC, and sequencing
- Strong organizational skills with the ability to handle multiple projects and high volumes of sample processing
- Properly use and maintain standard laboratory equipment
- Experience with long-read sequencing technologies, including ONT, is highly desirable
- Proficiency in nucleic acids extraction, quantitation, gel electrophoresis, PCRs, and related techniques is desired
- Experience with liquid handling and automated NGS pipelines is desired
- Clinical laboratory testing experience is desired
